چکیده

Formation of serine and pyruvate as intermediates in uric acid fermentation by Clostridium acidi-urici was suggested by Radin and Barker (1953), Sagers and Beck (1956), and Rabinowitz and B3arker (1956) to account for the labeling of acetate formed from C14-labele(d purines, and from the ability of cell suspensions and extracts to deaminate serine to pyruvate and ammonia. The present communication (lescribes the partial purification and some of the properties of an Lserine specific dehydrase from this organism. A preliminary report of this Work has been published (Benziman et al., 1959).

Intermediary metabolism of Clostridium acidi-urici. I. Formation of pyruvate from glycine.

Fermentation of purines by Clostridium acidiuirici was observed in early studies (Barker and Beck, 1941) to yield acetate, carbon dioxide, ammonia, glycine and formate. The formation of glycine and formate was confirmed more recently by Radin and Barker (1953), and Bradshaw and Beck (1953). Sagers and Beck (1956) have demonstrated, as was first suggested by Radin and Barker (1953), that L-serin...

Folate Coenzymes of Clostridium acidi-urici

(Z-)5,10-Methenyltetrahydropteroyltriglutamic acid was isolated from Clostridium acidi-urici. The procedure yielded approximately 25 mg of this stable derivative of the natural folate coenzyme per 100 g of wet cells. (I)-Tetrahydropteroyltriglutamate was synthesized by enzymic arsenolysis of the product. Procedures for the synthesis of (2)-10-[14C]formyltetrahydropteroyltriglutamate, (Z)-10-for...

The Use of 13C Nuclear Magnetic Resonance of Aromatic Amino Acid Residues to Determine the Midpoint Oxidation-Reduction Potential of Each Iron-Sulfur Cluster of Clostridium acidi-urici and Clostridium pasteurianum Ferredoxins*

13C NMR of the aromatic residues of Clostridium acidi-urici [Phe*]ferredoxin (a chemically modified ferredoxin in which a phenylalanyl residue replaces a tyrosyl residue) and Clostridium pasteurianum ferredoxin permits one to distinguish and probe each iron-sulfur (Fe&*) cluster neighboring the aromatic residues within each protein. This is because the ring carbon resonance shifts of the phenyl...

Crystal structure of Clostridium acidi-urici ferredoxin at 5-A resolution based on measurements of anomalous X-ray scattering at multiple wavelengths.

The crystal structure of Clostridium acidi-urici ferredoxin has been determined using multiple wavelength anomalous diffraction (MAD) techniques at 5.0-A resolution. The electron density map shows striking similarity to a map of Peptococcus aerogenes ferredoxin computed at the same resolution from the atomic coordinates reported by Adman et al. (Adman, E. T., Sieker, L. C., and Jensen, L. H. (1...

Tracer experiments on the mechanism of uric acid decomposition and acetic acid synthesis by Clostridium acidi-urici.

The relatively high yield of acetic acid from hypoxanthine is of considerable significance because a direct decomposition of the Ca chain in 1 mole of hypoxanthine cannot give more than 1 mole of a Cz compound. The observed yield of approximately 1.25 moles of acetic acid indicates that the formation of at least part of the acetic acid must involve a condensation either between two CB compounds...
